Update on Farxiga COVID-19 DARE-19 Phase III trial

AstraZeneca and Saint Luke's Mid America Heart Institute today announced high-level results of the primary analysis from the DARE-19 Phase III trial assessing the potential of Farxiga (dapagliflozin) to treat patients hospitalised with COVID-19 who are at risk of developing serious complications.

The trial did not achieve statistical significance for the primary endpoint of prevention measuring organ dysfunction and all-cause mortality, and the primary endpoint of recovery measuring a change in clinical status (from early recovery to death), at 30 days.

DARE-19 was the first Phase III trial to evaluate the safety and efficacy of a sodium-glucose co-transporter-2 (SGLT2) inhibitor in patients hospitalised with COVID-19 who also have risk factors for developing serious complications, including hypertension (HTN), type-2 diabetes (T2D), atherosclerotic cardiovascular disease (ASCVD), heart failure (HF) or chronic kidney disease (CKD) Stages 3-4.1,2 Cardiac, renal and metabolic comorbidities have been associated with poor outcomes and death in patients hospitalised with COVID-19.3,4

Tagrisso approved in China in early lung cancer

AstraZeneca's Tagrisso (osimertinib) has been approved in China for the adjuvant treatment of patients with early-stage (IB, II and IIIA) epidermal growth factor receptor-mutated (EGFRm) non-small cell lung cancer (NSCLC) after tumour resection with curative intent, with or without adjuvant chemotherapy as recommended by the patient's physician. Tagrisso is indicated for EGFRm patients whose tumours have exon 19 deletions or exon 21 (L858R) mutations.

US clearance of proposed acquisition of Alexion

AstraZeneca's proposed acquisition of Alexion Pharmaceuticals, Inc (Alexion) has achieved an important step toward completion, having cleared US Federal Trade Commission review. This follows the conclusion of the waiting period under the Hart-Scott-Rodino Antitrust Improvements Act.

The announcement follows competition clearances in Canada, Brazil, Russia and other countries globally, with a full list available on astrazeneca.com. Additional global regulatory clearances are pending, including but not limited to the UK, EU and Japan.

They will have a corrupt court in Brussels packed with commissioners who just so happen to be judges.

I think the Commission already has a lot of egg on its face. The legal action looks to me like an attempt to divert blame.

I don't know what the current figures are, but back in February it was said that only 21% of the AZN doses distributed to EU countries had been used!

According to the interesting (unofficial) French site, things are markedly better in France today - it would be interesting to see all the data. No doubt AZN's legal team will have the resources to explore this kind of thing.

Farxiga approved in the US for CKD

AstraZeneca's Farxiga (dapagliflozin), a sodium-glucose cotransporter 2 (SGLT2) inhibitor, has been approved in the US to reduce the risk of sustained estimated glomerular filtration rate (eGFR) decline, end-stage kidney disease (ESKD), cardiovascular (CV) death and hospitalisation for heart failure (hHF) in adults with chronic kidney disease (CKD) at risk of progression.

The approval by the Food and Drug Administration (FDA) was based on positive results from the DAPA-CKD Phase III trial. The decision follows the Priority Review designation granted by the FDA earlier this year.

A plus for AZN certainly; the incessant stream of vaccine related 'news' can distract attention from the main business but I am not so happy about the association with 'Never sell Shell'!

I have held both for over 10 years and as you can imagine, am much happier with AZN (XIRR 17.63%) than with RDSB (XIRR 5.58%).

Maxims can damage your wealth and over the past 3 years I have trimmed RDSB three times - putting the proceeds into HLMA, PHP and NG - so far, so good. It is now down to just 1.29% of my holdings; the kind of level where I should start thinking about adding or quitting.

By contrast I topped up AZN last month and it now forms 2.73% of my holdings. Not about to trim any time soon!

Thanks for your input. I get that one should never fall in love with a share. As for RDSB, I topped up my holdings four times last year, and have sat on my AZN shares, with no top ups, for years. Each to their own I guess.

Vaxzevria COVID-19 vaccine approved in Japan

AstraZeneca's COVID-19 vaccine, Vaxzevria (ChAdOx1-S [Recombinant]), formerly AZD1222, has been granted a special approval for emergency use in Japan for active immunisation of individuals aged 18 years and older, to prevent COVID-19 caused by SARS-CoV-2.

The Japanese Ministry of Health, Labour and Welfare granted the approval based on positive Phase III efficacy and safety data from the Oxford University-led clinical trial programme in the UK, Brazil and South Africa, and a Phase I/II trial in Japan.

AstraZeneca appoints new Chief Financial Officer

AstraZeneca PLC announced today that the Board has appointed Aradhana Sarin as an Executive Director and Chief Financial Officer, conditional upon closing of AstraZeneca's acquisition of Alexion Pharmaceuticals, Inc. (Closing) and effective on the later of 1 August 2021 or the date of Closing, which is expected to take place in the third quarter of 2021, subject to receipt of certain regulatory approvals.

Dr. Sarin is currently Executive Vice-President, Chief Financial Officer of Alexion. Further biographical details are set out below. Dr. Sarin will relocate from the US and be based in the UK. She will report to AstraZeneca's Chief Executive Officer, Pascal Soriot.

Lynparza reduced recurrence risk in breast cancer

Results from the OlympiA Phase III trial showed AstraZeneca and MSD's Lynparza (olaparib) demonstrated a statistically significant and clinically meaningful improvement in invasive disease-free survival (iDFS) versus placebo in the adjuvant treatment of patients with germline BRCA-mutated (gBRCAm) high-risk human epidermal growth factor receptor 2 (HER2)-negative early breast cancer.

The results will be presented during the plenary session of the 2021 American Society of Clinical Oncology (ASCO) Annual Meeting on 6 June 2021 (abstract LBA#1) and were published today in The New England Journal of Medicine.

Calquence head-to-head results versus ibrutinib

Final results from the head-to-head ELEVATE-RR Phase III trial of AstraZeneca's Calquence (acalabrutinib) demonstrated non-inferior progression-free survival (PFS) and statistically significantly fewer events of atrial fibrillation versus ibrutinib in adults with previously treated chronic lymphocytic leukaemia (CLL), the most common type of leukaemia in adults.1

Update on AZD7442 STORM CHASER trial

AstraZeneca today announced results from the STORM CHASER trial assessing the safety and efficacy of AZD7442, a long-acting antibody (LAAB) combination, for the prevention of symptomatic COVID-19 in participants recently exposed to the SARS-CoV-2 virus. The trial did not meet the primary endpoint of post-exposure prevention of symptomatic COVID-19 with AZD7442 compared to placebo.

Orpathys approved in China for lung cancer

AstraZeneca and HUTCHMED's Orpathys (savolitinib) has been granted conditional approval in China to treat patients with non-small cell lung cancer (NSCLC) with MET exon 14 skipping alterations who have progressed following prior systemic therapy or are unable to receive chemotherapy.

This approval follows a priority review designation by the Center for Drug Evaluation of China's National Medical Products Administration (NMPA) and marks the first global regulatory approval for the oral, potent, and highly selective MET tyrosine kinase inhibitor (TKI).

More than a third of the world's lung cancer patients are in China and, among those with NSCLC, approximately 2-3% have tumours with MET exon 14 skipping alterations, a targetable mutation in the MET gene.1-3 This mutation is more common (13-22%) among patients with pulmonary sarcomatoid carcinoma (PSC), a rare and aggressive subtype of NSCLC usually resistant to chemotherapy.1,4
